What is the optimal bullet diameter for the Shiloh 45's?
Can a 45-70 be fired out of a Shiloh 45-110?

0.458", per Kirk (owner and builder). 0.459"-0.460" will also work.
.45-70 can physically be fired from.45-110, but I would expect very little in the area of accuracy, because there may be significant blobby and gas cutting, bullet will expand significantly, then jump for ~3/4" before engaging the rifling, and significantly contract as it does so. Definitely not a recommended practice!

458 diameter if you're talking greasers.
Yes a 45-70 can be fired in a 110 chamber, accuracy will be crap, leading in the front of the chamber and the throat will be a nightmare to get cleared out, and done enough times could possibly gas cut your chamber.
Buy the correct brass, and 1f powder by the case.
