How are the MVA Buffalo Soule sites set (zeroed) at the factory before the gun is shipped? What assumptions are made as to the initial ballistics for a 45-70 round and at what point is the calculated Zero.

I'm not real sure what you're asking here. We mount the sights perpendicular with the rifle. It makes no difference what caliber they are going on.

I guess they are the same as any other optic in that you set the Vernier to 0 shoot a group at 100 yrds ( or whatever distance you choose), and make a ballistic table from there.

No, your Vernier will be a number at a 100 yards, depending on caliber. There is no set number, it may be 25 points depending on front sight height. then usually with black powder you will be close at 200 yards with a ten point move in elevation and so on.

Watch the video above.
It's titled "Shiloh Rifle from Foundry to Finish", skip to the 9:10 second mark & pay attention.
Neat trick. Gotta remember that one

I'm more interested in the how to's of getting the staff perpendicular to the bore. How that is quickly & easily achieved, patiently waiting the newly produced video

I get it. Thanks for all your good advice. I rely on the Sharps people to set the sights perpendicular, then I can go from there and develop my sight tables and load tuneing for the rifle. Really looking forward to this new/old shooting experience with the help from a little applied Physics and tuning from you all's expirience as I go.
